Preacher acompanha a história de Jesse Custer, um ex-pastor texano em conflito que acaba possuído por uma força espiritual fugitiva do céu chamada Genesis, a qual confere a ele o poder de fazer com que qualquer um o obedeça. Acompanhado de sua ex-namorada Tulipa e o vampiro irlandês Cassidy, Custer vai atrás de Deus a fim de encontrar as respostas que procura.
